Endothelin receptors and responses ::
There are two types of ET receptors -ETA and ETB Vascular smooth muscles contain only ETA receptors Other tissues like brain kidney intestine adrenal gland contain large number of ETB receptors Endothelium have least ETB receptors Main functions of ETA receptors are vascular smooth muscle contraction bronchoconstriction and aldosterone secretion Stimulation of ETB receptor produces vasodilatation and inhibition of ex-vivo platelet aggregation ET3 produces potent ETA receptor stimulation All three types are otherwise equieffective on ETA and ETB receptors
Endothelin Antagonists ::
1.ET A Receptor Antagonist
Steroid derivative from bayberry
Butenolide derivative

2.ETB Receptor Antagonist
BQ-788
3.Non -Selective Endothelin Antagonists
Bosentan
RO-46-2005
Indanecarboxylic acid derivatives
Cyclic Pentapeptide
Bosentan ::
Bosentan is the first oral endothelin antagonist approved for treatment of Primary pulmonary hypertension and Pulmonary arterial hypertension (PAH) Primary pulmonary hypertension is defined as abnormally high blood pressure in the arteries between the heart and the lungs
